Research Abstract

1.マボヤ卵黄膜蛋白質VC70のユビキチンリガーゼの探索:すでに、マボヤ精子放出液からVC70蛋白質をユビキチン化する分子量700kDaのユビキチン化酵素を単離し、さらに、そのユビキチン化活性を促進するユビキチンリガーゼE3様酵素を、ユビキチン化サイトLys234を含むVC70ペプチドを固定化したカラムを用いたクロマトグラフィーにより単離した。しかし、得られた蛋白質量が微量のために生化学的解析が不可能であったので、上記のVC70ペプチドをベイトに用いて酵母two-hybridスクリーニングを行い、2つのVC70ペプチド結合蛋白質を得た。一つは、UBAドメイン含有蛋白質で、これはVC70蛋白質が酵母内でユビキチン化され、そのポリユビキチン鎖を介して結合蛋白質として得られたと考えられる。一方、マボヤの近縁種であるカタユウレイボヤのESTデータベース検索の結果、もうひとつのクローンは全長ではなく、N末端配列を欠失したものであると推測される。この全長をクローニングし、VC70蛋白質のE3酵素である可能性や受精における役割を明らかにすることが今後の課題である。2.胚特異的ユビキチンレセプターの機能解析:アフリカツメガエルの胚特異的ユビキチンレセプターRpn10cが抗アポトーシス因子Scytheと特異的に結合する。Rpn10cとの結合に関与するScytheのN末端領域を解析し、それがタンデムに連結した4つのユビキチン様ドメインからなり、少なくとも2つのドメインの存在がRpn10cとの結合に必要であることを明らかにした。また、そのドメインがアポトーシス制御に必要であることを明らかにした。Rpn10cを含む胚特異的プロテアソームがScytheを介してどのように発生過程におけるアポトーシスを制御しているかを明らかにすることが今後の課題である。
