Research Abstract

オートファジーの活性をモニターしうるGFP-LC3(酵母Atg8ホモログ)トランスジェニックマウスの解析によって、マウスにおいては出生直後に全身で非常に活発なオートファジーが誘導されることを明らかにした。そこで、この生直後のオートファジーの栄養学的な意義をAtg5ノックアウトマウスを用いて検討した。APG5-/-産仔は出生後24時間以内に全例死亡する。この主な理由はミルクの摂取が非常に悪いためであった。しかし帝王切開後絶食という栄養条件をそろえた条件でも、野生型が平均21時間生存するところ、KOマウスは平均12時間で死亡した。このKOマウスの早期の死亡は経胃管栄養によって部分的に抑制された。さらにKOマウスでは血中および組織内のアミノ酸濃度が有意に低下しており、細胞内は低エネルギー状態にあった。このことから、経胎盤栄養が途絶えることによる生直後の飢餓を凌ぐためには、一過性に誘導されるオートファジーによるアミノ酸供給が重要であることが示唆された。また、このAtg5ノックアウトマウスに由来する線維芽細胞を用いて、オートファジーが細胞内侵入細菌の分解や、アポトーシス抑制時の細胞しなどの多彩な生理機能を有していることを明らかにした。
一方、メタノール資化性酵母を用いたApgの解析においては、ペキソファジー特異的に必要な新分子Atg24を同定した。Atg24はイノシトール3リン酸結合部位であるPXドメインを持つが、MIPAと呼ばれる新生膜の形成やペキソファゴソーム形成には不要で、ペキソファゴソームと液胞の融合に必要であった。今後は、PpAtg8を用いて新生膜の形成・液胞膜の変形などを追跡し、様々なAtg分子の機能・時空制御機構について明らかにしていく予定である。
