Research Abstract

可溶性グアニル酸シクラーゼの研究に関しては、ヘム結合ドメインの大量発現系および精製法を確立したが、結晶化条件を最適化するまでには至らなかった。
ヘムオキシゲナーゼ(HO)の反応機構については詳細に迫ることができた。今年度の実績は以下の通りである。
ヘムオキシゲナーゼの活性中心には効率的なプロトン供給チャネルが存在し、特にパーオキシ型活性種の生成に必要なプロトン移動は、酵素の構造緩和が起きえない4Kにおいてさえ速やかに進行することを明らかにした。同様なプロトン供給チャネルを有する他の酵素と比べても、ヘムオキシゲナーゼにおけるプロトン移動は非常に速く、ヘム分解という特異な反応に必須の要素ではないかと考えられる。なお、本研究は酵素中のプロトン移動を液体ヘリウム温度で観測した初めての例である。
ベルドヘム開環はヘム分解の律速段階にあたる重要な反応であるが、その反応機構などはほとんどわかっていない。詳細な反応解析の結果、従来から知られていた酸素のみでなく、過酸化水素によってもベルドヘム開環が進行することを見いだした。いずれの反応においても反応中間体の直接観測に初めて成功し、反応機構の解明に向けて重要な知見が得られた。また、過酸化水素による反応は生体内でも起きうる可能性があり、ヘムオキシゲナーゼは二つの機構を使い分けによって、酸化ストレスに迅速に応答すると考えられる。
ほ乳類のヘムオキシゲナーゼには二種類のアイソザイム(HO-1,HO-2)が存在する。結晶構造に基づいた薬剤デザインにより、一方のアイソザイムのみに作用する阻害剤の開発を進めている。現在までに、薬剤候補分子の骨格デザインを終え、HO-1に対して約15倍の選択性を示す分子を得ている。
また、4つの反応中間体や基質が結合していないヘムオキシゲナーゼの原子レベルの結晶構造解析に成功し、反応を制御する構造的な要因を明らかにした。
