Research Abstract

肝細胞増殖因子(HGF)受容体(c-Met)を介するがん細胞の増殖制御におけるシグナル伝達機構について、HGFによる肝がん細胞株HepG2の増殖抑制作用における細胞周期制御因子であるcdkインヒビターp16の役割を解析し、以下の結果を得た。
1.p16のsiRNAにより細胞内のp16のHGFによる発現誘導を阻害した時の細胞周期について解析したところ、p16のsiRNAの発現によりHGFにより抑制されたBrdUのDNAへの取り込みが回復し、またHGFによりG1期で停止した細胞周期の回復が認められた。これらの結果は、HGFにより発現が誘導されたp16がHGFによる細胞周期のG1期での停止に必要であることを示している。
2.HepG2細胞では、細胞周期を制御するキナーゼであるcdk2の活性低下が、HGFによる細胞周期の停止を誘導すると考えられている。HGFによるcdk2活性の低下とp16の発現誘導の関係を解析したところ、両者はよく相関していることが明らかになった。またHGFにより低下したcdk2活性は、p16のsiRNAの発現により回復した。これらの結果は、HGFによるp16の発現誘導が、細胞周期の停止を導くcdk2活性の低下に必要であることを示している。
3.cdk2の活性はp21やp27などのcdkインヒビターにより阻害される。p16,p21,p27とcdk4,cdk2との結合について解析したところ、HGFによりp16とcdk4との結合が増加し、またp21あるいはp27とcdk4との結合が低下し、cdk2との結合が増加した。またp16のsiRNAの発現によりこれらの結合の変化は見られなくなった。これらの結果は、HGFによるp16の発現誘導がp21およびp27のcdk4からcdk2への移行に必要であり、その結果cdk2の活性の低下が導かれることを示している。
