Research Abstract

1 ras癌遺伝子依存性の発癌におけるRas/Rap1標的蛋白質ホスホリパーゼCε(PLCε)の機能解析のため、遺伝子ノックアウトマウスを作製しDMBAをイニシエータ、TPAをプロモータとした二段階皮膚化学発癌実験を行なった。ノックアウトマウスでは、野生型マウスに比べパピローマの発生時期が遅延し発生個数が約4分の1に減少した。更に、扁平上皮癌へのプログレッションが全く起こらなかった。発生した全腫瘍において、H-ras遺伝子の61番コドンに活性型変異が見られた。この結果は、PLCεがras癌遺伝子依存性の個体レベルでの発癌に重要な役割を持つことを示した。また、ノックアウトマウスでは、TPA処理により誘導される皮膚基底層細胞の増殖と肥厚が起こらず、PLCεがホルボールエステル下流のシグナル伝達に重要な役割を持つ事を示した。これらの結果は、PLCεが抗癌剤開発の新しい分子標的になることを示唆した。
2 PLCεノックアウトマウスは、先天性心臓半月弁膜症に起因する心室拡張を示した。半月弁の弁葉の肥厚と形態異常を引き起こす分子機構としては、PLCεが上皮増殖因子(EGF)受容体-Ras経路の下流で、骨形成因子受容体下流でのSmad1/5/8の活性化の抑制に関わると推測した。
3 Ras類似蛋白質M-Rasの高次構造解析を通じて、GTP結合型Rasの中に標的蛋白質との結合能力を有する型(state2)と有しない型(state1)が相互変換が可能な状態で存在する事を見いだし、state1の高次構造の決定に世界で初めて成功した。State1は、GTPのγ-リン酸基とRasのセリン-35残基側鎖との結合が解離しており、その結果生じたポケットに嵌入することによりstate1を安定化する物質がRasの機能阻害を通して新しい抗癌剤として使える可能性を示唆した。
