Research Abstract

アポトーシスのシグナル伝達機構ならびに細胞増殖のシグナル伝達機構を標的とした放射線増感の可能性についてさらに検討するとともに、Hsp90シャペロンコンプレックスやヒストン脱アセチル化など放射線感受性増強のための新たな分子標的の探索とその機構の解明を行い、以下の結論を得た。1)ホルモン感受性前立腺癌においてアンドロゲン受容体を介した増殖刺激は放射線感受性の低下を誘導するが、Hsp90シャペロンコンプレックスの阻害により相乗的な放射線増感効果が認められ、Hsp90シャペロンコンプレックスはホルモン感受性前立腺癌においても放射線応答に重要な役割を果たしていた。2)ヒストン脱アセチル化の阻害はp53非依存性のアポトーシスに加えて、生存シグナル伝達という放射線抵抗性に働くひとつのファクターを抑制できることが明らかとなった。3)上皮細胞増殖因子受容体であるEGFRならびにHER2/neuの2つの受容体からのシグナルを同時に阻害することで相乗的な放射線感受性の増強が得られるかどうかをEGFRならびにHER2/neuの2つの受容体を発現するA431細胞株を用いて検索したところ、ZD1839はEGFRのりん酸化を抑制するだけでなく、HER2/neuのりん酸化をも抑制した。また、EGFRとHER2/neuを同時阻害することによって相乗的な放射線感受性の増強が認められ、受容体の複数阻害によって効率的な放射線増感をもたらす可能性が明らかとなった。4)放射線照射により異なる放射線感受性を有するPNET細胞株を樹立し、放射線応答の違いに関して遺伝子発現の相違をマイクロアレイで検討すると、通常状態で抵抗性株に発現の高い遺伝子として11種類の遺伝子が、抵抗性株に発現の低い遺伝子として11種類の遺伝子が明らかとなった。
