Research Abstract

超音波は癌の診断に幅広く用いられているが、高密度焦点式超音波を用いた癌治療等、治療への応用が期待されている。物理的エネルギーの癌治療への応用と最適化を目指して、超音波、放射線および温熱を対象に作用を比較し研究を進めてきた。本年は特にアポトーシスに関連した細胞内活性酸素の役割の解明を目的とした。超音波で顕著に発現誘導されるヘムオキシゲナーゼ-1の遺伝子および蛋白質の発現を、照射後に添加したヒドロキシル(OH)ラジカル除去剤が効率的に抑制することを明らかにした。放射線および温熱アポトーシスに対する抗酸化剤の影響についても検討し、アポトーシス抑制効果は単純にOHラジカルおよびスーパーオキシドの消去活性に依存しないことが判明した。温熱によって発現が変動する遺伝子をマイクロアレイ解析で調べたところ、時間変化により4つの遺伝子群に分類できた。最も発現変化の大きかった遺伝子はc-junであった。同蛋白質およびmRNAの発現は抗酸化剤で抑制され、本遺伝子は温熱による細胞内酸化ストレスの指標となることを示した。スピン捕捉剤であるPBNは酸化ストレスにより一酸化窒素(NO)を遊離することから、温熱との併用を検討したところ、NOおよびONOOによるアポトーシス増強作用を新たに見いだした。超音波による遺伝子導入については超音波造影剤YM454とLevovistについて前立腺癌細胞を用いて比較し、同じ生存率条件では前者が約50倍優れることが判明した。超音波は遺伝子導入、アポトーシス誘導および遺伝子発現の制御に利用でき、将来の癌治療に有用と思われる。
