Research Abstract

我々は昨年度までに、緑茶や十字科野菜の摂取が多い患者では、胃がんにおけるホメオプロテインCDX2遺伝子のメチル化頻度が低いことを報告し、Epigenetic epidemiology(エピジェネティック疫学)の概念を確立した。今年度はさらに研究を進め以下の研究結果を得た。
骨形成因子(BMP)-2の胃がん細胞における発現を調べた結果、プロモーター領域のメチル化が発現抑制の原因であった。原発胃がんの解析でも、メチル化が発現抑制に重要だった。BMP-2のメチル化頻度は未分化型で高く、BMP-2が胃がん、特に未分化型で発症に関連している可能性が示唆された。次に、がんで異常メチル化の多い染色体3p21に着目し、その領域にあるカルシウムチャンネル構成因子の一つをコードするCACNA2D3遺伝子のメチル化を解析した。その結果、やはり未分化型胃がんでメチル化頻度が高かった。
DNAメチルトランスフェラーゼ(DNMT)3Aと3Bは、メチル化に必須であり、それらの遺伝子の発現調節機構を調べて、Spファミリー因子が重要であることを明らかにした。胃の幽門腺特異的ムチンMUC6をコードする遺伝子の発現調節機構を解析した結果、Spファミリー因子とNF-κBが正に調節していた。
CDX2の発現を胆嚢がんで解析して、37%で陽性であった。CDX2とMUC2の発現は有意に相関しており、胆嚢がんでも腸型形質を示すがんがあって、CDX2が関連している可能性が示唆された。
