Research Abstract |
Although it has been reported that bone marrow cell could be introduced to cardiomyocyte by the stimulation of 5-azacytidine, its efficacy has been low and many points must be resolved were remained. We studied the new method for introduction from bone marrow cell to cardiomyocyte in stead of 5-azacytidine. Method: Bone marrow cells were cultured on the silicone sheet. And then the sheet was periodically stretched at 60 Hz. The change of cell figure was checked everyday. And the expression of N-cadherin was examined because N-cadherin is specifically up-regulated in the gap junction of cardiomyocytes. Results: Cultured bone marrow cells stretched periodically changed its shape and axis. Cells formed the connection between cells and became one theread like cell line. At last these threads formed network of cells which is very resembled to heart tissue. Immunostaining by anti-N-cadherin staining showed that gap junctions were formed between cells. Conclusions: Cardiomyocyte was introduced by the stimulus resembling the environment in the heart tissue.
