Research Abstract

ポリコーム遺伝子群はショウジョウバエの遺伝学的解析から発見されたが、我々はそれらの哺乳動物相同遺伝子を単離し、遺伝子欠損マウスを作製することによって、哺乳動物の発生制御にはポリコーム遺伝子群を介した遺伝子発現維持の機構が重要な役割を果していることを明らかにしてきた。興味深いことに、ポリコーム遺伝子群による遺伝子発現維持機構が、造血幹細胞の自己複製、血球分化や白血病に関わるだけでなく、心臓形態形成、さらに心筋維持の機構にも重要な役割を果していることを明らかにすることができた。造血制御に関しては臨床研究をも平行して行い、また心筋の肥大と維持の機構に関してもシグナル伝達に関する詳細な解析を行うことができた。これらの一連の解析によって、その概念すら新規な遺伝子発現維持の機構がどのような高次生命現象、さらにヒト疾患に関わっているかを明らかにするための基盤を形成することが出来た。これらの知見を基に、今後、クロマチンの高次構造を介した転写制御の観点からその詳細な分子機構の解析を更に進めるだけでなく、発生が完成し、維持される機構、特に幹細胞システムの機能維持のメカニズム解明へと研究を展開し、今後の再生医学の発展に貢献したいと考えている。
