Research Abstract |
Systematics and biogeography of Chinese and Korean bitterlings were studied. In 2000 and 2001, we went to Shanghai City, Hainan and Hunan Provinces and Guangxi Zhuang Autonomous Region in China, and Gyonggi, Chungchongbuk, Chungchongnam, Gyongsangbuk, Jollabuk, Jollanam Provinces in Korea, and Taiwan, where we collected samples for classification, karyotypes and DNA experiments, with Chinese and Korean investigators. As the results of the Expedition from 2000 to 2001, following points were found : 1) Rhodeus sp. collected from the Namhan River system, Korea, was reported as a new bitterling, Rhodeus pseudosericeus. 2) Developments of eggs and larvae of Rhodeus pseudosericeus collected in Korea and Rhodeus spinalis collected in Hainan Province, China, were studied. olk-sac is dented in the hatched R. pseudosericeus larva, and the anterior part of york-sac is plump in the hatched R. spinalis larva. 3) The intense C-band in the middle region of the long arm in the subtelocentric chromosome was observed in Rhodeus ocellatus. The number of chromosome pairs with this C-band varied from 0 to 3 depending on the geographic site of collection. 4) The chromosomal polymorphism of 2n=44 and 2n=45 was recognized in males of Acheilognathus rhombeus in Korea. 5) The complete nucleotide sequence of Rhodeus ocellatus kurumeus mitochondrial genome was determined. A circular mitochondrial genome from the fish was 16,674 bp in length, and showed the same organization as generalized vertebrate mitochondrial genomes. 6) Sequence analysis of cytochrome b region in mitochondrial DNA (1,141 bp in total) was performed to infer the phylogenetic interrelationships of bitterlings, with 37 species or subspecies in China, Korea, Taiwan and Japan. In all phylogenetic trees, bitterlings are mainly composed of two clades, Acheilognathus clade and Tanakia-Rhodeus clade. Less
