Research Abstract

1. Findings on an action mechanism of PACAP of the mediator of NANC relaxation in the rat distal colon are as follows.
(1) PACAP induces relaxation of the muscle via a decrease in intracellular Ca2+ ions by activation of PAC1 receptors which, in turn, opening of small conductance Ca2+-activated K+ (SK) channels.
(2) SK3 was found to be expressed in the rat distal colon. SK3 was localized around the myenteric plexus, but not in the smooth muscle.
(3) From the immuno-electronmicroscopy studies, it was suggested that SK3 is associated with interstitial Cajal cells and has an important role on the inhibitory transmission.
2. Findings on a role of orexin on an inhibitory transmission in the mouse small intestine are as follows.
(1) Exogenously added orexin induced relaxation followed by contraction which is induced via muscarinic receptors in longitudinal muscle of mouse small intestine.
(2) It was shown that about a half of neurogenic relaxant response was mediated by nitric oxide which was produced by orexin.
3. An intracellular mechanism of relaxation of the rat proximal colon by nitric oxide was suggested: nitric oxide activates SERCA-2 and decreases intracellular Ca2+ ions.
4. By studying roles of Ca2+ store in SR and inward rectifiered K+ channels on the motility of the gastrointestinal tract in addition to these studies, papers listed below were published.
