Research Abstract

The expression of NF-Y protein was detected in the cancer tissues of the breast, prostate, and stomach using a catalyzed signal amplification system for immunohistochemistry. Especially, gastric cancer cells showed a higher ratio of NF-Y expression in their nuclei, and also those of normal gastric epithelial in proliferative phase. The immunolocalization of proliferating nuclear antigen and MIB-1 were very consist with that of NF-Y. This suggested that the NF-Y is a potential new maker for gastric cancer cell proliferative activity. The cdc2 expression, which is regulated by NF-Y, was not detected in the cancer cells, possibly due to the antibody specificity we used for immunostain.
The cdc2 gene promoter activity in cultured human gastric cancer cells was reduced by dominant negative NF-Y.
Taken together, we showed that the NF-Y regulates the cdc2 gene promoter activity and that its expression is high in gastric cancer cell.
