The role of p14/ARF in the development of adult T cell leukemia/lymphoma (ATLL) was studied. p14/ARF DNA was deleted in a cell line, OKM3T, among nine ATLL related cell lines (MT1, MT2, HUT102, OKM3T, F6T, K3T, Oh13T, S1T, and SU9T01). The gene was not expressed in OKM3T and weakly expressed in S1T by RT-PCR analysis. We are investigating the methylation status in the promoter region of p14/ARF in S1T. p14/ARF and MDM2 proteins were detected in one and two of 11 lymphnodes derived from primary ATLL patients by immunohistochemical staining, respectively. P53 protein positive cells were 7.51% (mean) in ATLL cells, which is not significantly different compared with other lymphoma cells. Protein expression of both MDM2 and p53 were correlated neither with the expression of p14/ARF nor grade of malignancy in lymphoma. The regulation mechanisms of p14/ARF, MDM2, and p53 as well as contribution of these genes to development of ATLL should be further studied.
