Research Abstract |
We have observed that elevation of serum levels of IL-18 in some cases of aplastic anemia. Here, we have tried to clarify the role of IL-18 in bone marrow failure in mice. Treatment ofPropionibacterium acnes (P. acnes) and LPS in mice is a model forendotoxin-shock and fulminant hepatitis, in serum of this model mouse IL-18 was first discovered. We analysed bone marrow of this mouse and found massive bone marrow apoptosis and hematopoietic failure. Pretreatment ofP. acnes induced Fas (CD95) expression on c-kit+ hematopoietic progenitor cells and suscepitbilty for Fas-mediated suppression of hematopoietic colony formation. Subsequent treatment of LPS induce Fas ligand in bone marrow mononuclear cells and massive apoptosis of marrow cells. This marrow failure was transient and recovered after 48 hours. Pretreatment of anti-IL18 antibody prevented the bone marrow apoptosis. These findings suggests that IL-18 is involved in Fas-mediated massive marrow apoptosis though Thl pathway in endotxin schok mice.
