Research Abstract |
The blood plasma and seminal plasma involved adenyl purins. In the former, ATP occupied more than 50% and cAMP was under-detectable. On the other hand, adenosine occupied 57% in the latter and ATP was found to be less than 1.0%. The composition of adenyl purines in two body fluids were quite different each other. Adenyl purine metabolisms in the blood plasma and seminal plasma were compared each other. When added ATP in the seminal plasma, ADP, AMP, adenosine were increased as the time proceeded. This fact suggested that ATP was degraded by a de-phosphorylation. On the other hand, the adding Atp in the blood plasma readily disappeared by degrading the purine structure. To observe the effects of ATP on sperm acrosome raction, contamination of the seminal plasma could not be neglected, the sperm was purified by means of a linear gradient centrifugation in Percoll with an inner-column. After centrifugation, residual seminal plasma acid phosphatase activity was decreased to be 0.003% of the row semen. The acrosome reaction (A) was evaluated with FITC-concanavalin A/PI fluorescent staings. ATP accelerated the induction of A, and the ATP receptor antagonists disappeared the effect. UTP gave the strongest effects on A, and it was suppressed by PKC inhibitors and Ca channell blockers. On the other hand. Adenosine suppressed the induction of A. The above results suggested that adeny purines might play important roles for the regulations of A in the male and female reproductive tracts.
