Research Abstract

1. First of all, we have picked up 16 kinds of genes (Evi-1 protein, SNF/SW12-related protein, sucrase-isomaltase, homeodomain protein (OG12), Human TNF-related apotposis inducing ligand TRAIL, PLD1a, chromosome-associated polypeptide-C, TSA2004, cholineesterase, glucose transporter-like protein, four transmembrane domain protein, deubiquitinating enzyme, htra-2 beta, translocation protein-1, apolipoprotein D precursor, PIK3CA) located around 3q26.2 locus by searching genome data base and constructed PCR primer pairs derived from their cDNA sequences. Secondly, we have investigated expression of their genes by RT-PCR method on nine human ovarian cancer cell lines (HAC-2, OMC-3, HOC-21, HOC-1, HUOKA II, SHIN-3, HMOA, HUOA, OVCAR-3). Finally, we have confirmed each cell line had individual gene expression pattern and the expression pattern had no correlation with histological subtype of the original tumor. 2. We have investigated the mutations of beta-catenin and gamma-catenin genes on human ovarian cancer cell lines by RT-PCR and SSCP and have found no homozygous deletion and point mutation.
