Research Abstract |
Recent studies have been saised about the possibility that one of estrogenic chemicals, bisphenol A (BPA) might be leached out of dental sealants. This study aimed to identify and quantify BPA and other components released from sealants and dental polycarbonate materials in vitro. The sealants tested were not shown to have released BPA analyzed by a liquid chromatography. The three dental polycarbonate materials contained the amounts of 28.8 to 203 μg of BPA pre gram of pellet. But the content of BPA of dental PC material extracted with PBS(-) was less than μg level per gram dental PC material. The estrogen-sensitive MCF-7 breast canser cells was a useful tool that may estimate an estrogenic chemicals (environmental estrogens, xenoestrogens). When incubated with MCF-7 cultured with dextran-charcoal treated FCS and phenol red-free medium, BPA, 17 β-estradiol, diethylstilbestrol (DES) and ethynylestradiol promoted cell proliferation that was assayed the DNA content by DABA method, and induced the expression of progesterone receptor mRNA that was analyzed by RT-PCR. The extracts of dental sealants and dental PC materials with PBD(-) were not affected the proliferation and the expression of progesterone receptor of MCF-7.
