Research Abstract |
Purpose : Maternal environmental factors have much influence on the occurrence of cleft lip and/or palate. As incidence of cleft lip and/or palate in CLP-prone A/J strain mouse decreases by administrating estradiol, it is suggested that various hormones affect facial generation. As environmental hormone acts like female hormone, its influence to human body is concerned. Therefore, we studied about the influence of environmental hormone to cleft lip and/or palate. Method : We used A/J strain mouse (Japan SLC) for experiment. As incidence of cleft lip and/or palate decreases to a minimum when estrogen is administered on the 9^<th> gestation day, BisPhenolA was administered on the 9_<th> gestation day. Amount of BisPhenolA administered was 0, 0.1, 10mg/kg each. Incidence of cleft lip and/or palate was investigated on the 18^<th> gestation day. % in 0.1mg/kg group, 4.91% in 1mg/kg group and 7.31% in 10mg/kg group. Though incidence of cleft lip and/or palate decreased in the group administered 0.01mg/kg BisPhenolA and 0.1mg/kg group, incidence increased as amount increased. Discussion : Molecular of BisPhenolA is so small that it reaches fetus through placenta. It combines with estrogen receptor and acts like female hormone. As cephalo neural crest cell has an important role in maxillofacial generation, it is well known that interruption of its move or growth causes congenital anomalies like cleft lip and/or palate. It was considered that move of neural crest cell is almost completed in the BisPhenolA administered fetus at 9^<th> day, BisPhenolA acted to the cell and promoted its proliferation. Also, environmental hormone in large amounts induces congenital anomalies like cleft lip and/or palate. Therefore, it suppressed incidence of cleft lip and/or palate for 0.1mg/kg administered group, but it showed teratogenicity for 1mg/kg and 10mg/kg groups and incidence of cleft lip and/or palate increased.
