Research Abstract

Many oral cancer cell lines have been established, their invasion and metastasis are analyzed. But, establishment of adenoid cystic carcinoma cell lines are very little because the cell line is slow growth. This study was designed to examine the establishment of in vivo selection lung metastasis animal model by homotransplantations of human adenoid cystic carcinoma in nude mice, of high lung metastasis cell line, and expression of matrix metalloproteinase. The results obtained were as follows.
1) We transplanted the oral floor human adenoid cystic carcinoma into the oral floor of the nude mice. But naked eye or histopathologically, tumor of oral floor and lung metastasis were not revealed 4 months after the transplantation. We have been revealed the micro invasive oral floor tumor in the nude mice 1 year after the transplantation, but the lung metastasis was not revealed. Early lung metastasis cannot reveal by the naked eye, and so the lung tissue was transplanted into the subcutaneous tissue of the nude mice, we have been observed growth of the tumor. In the present, the natural neck metastasis tumor is observed by serially transplantation and spontaneously metastasises to the lung when transplanted in nude mice. For the natural neck metastasis and lung metastasis tumor were undifferentiated carcinoma histopathologically, differentiation of the tumor are performed
2) As in the above because the lung metastasis tumor line are creating, we are not establish of adenoid cystic carcinoma cell line.
3) We also have no data expression of matrix metalloproteinase.
In this experiment, the tumor is slow growth above expecting and lung metastasis is not shown at present, furthermore continuous research of several years is necessary to exert the result.
