Research Abstract

We examined Bidens parviflora with NO production inhibitory effects and prostaglandin E_2 (PGE_2) inhibitory effect toward the macrophage secretions which had a relation in antiallergy and chronic inflammation. Chinese folk medicine, Bidens parviflora (Compositae) has been used as a traditional antipyretic and anti-rheumatic medicine. Sixty-one compounds isolated from this whole plant were examined for anti-allergic actively toward nitric oxide (NO) production by a murine macrophage-like cell line, RAW264.7, activated with lipopolysaccharide (LPS) and interferon-γ(IFN-γ) and histamine release from rat peritoneal mast cells in vitro. We isolated five new polyacetylene glucosides, twenty-five flavonoides containing two new compounds, a new dimeric caffeic acid derivative, two new caffeic acid esters, two caffeic acid derivatives, two cinnamic acid derivatives, two new neolignans, a cyclopentene glycoside having ten carbon chain, eight quinic acid esters, a benzyl alcohol glycoside, a naphthopyrone glycoside, carotinoids, a diterpene, sterols and myo-inositol.
Among five polyacetilene glucosides, four exhibited strong inhibitory activities toward NO production and, about effect to give to NO product performance of a macrophage of these amalgam, restraint was recognized as a result of having examined action of an examination drug at having been revitalized with IFN-γ and LPS. In the flavonoids, NO product inhibitory activities were recognized by flavones, flavonols and aurones which had two hydroxyl groups to 3', 4' -position. In addition, NO product inhibitory activities were recognized, by a caffeic acid dimer, a caffeic acid ester of methyl quinic acid. We investigated the inhibitory effects of isolated compounds on prostaglandin (PG) E_2 production in LPS-activated macrophages. About sucrose p-hydroxylcinnamic acid esters and caffeic acid derivatives, PGE_2 production inhibitory activities were recognized by several compounds.
