Rat was trained for the lever-press for food reward. Striatal dopamine (DA) and serotonin (5-HT) release was measured every 250 ms. After cue (tone), rat could get a pellet if the lever was pressed. Tone was applied every 7 s, totally 20 times in a session. Six sessions were performed every 30 min. After 20-30 experimental days, signal currents were analyzed. DA current decreased temporally just after the tone stimulation, but it increased until around the lever-press. Then, DA current decreased again. Conversely, 5-HT current increased temporally after the tone stimulation, but soon it decreased. Then, 5-HT current seemed to return to the basal levels. PCPA (5-HT synthesis inhibitor) treatment diminished the temporal suppression of DA current and the increase around at the lever-press a-MPT (DA synthesis inhibitor) did not affect 5-HT release From these findings, it was concluded that the temporal suppression of DA release is caused by inhibitory 5-HT neuron, and that DA increase around at the lever-press may be partly facilitated by 5-HT neuron.
