アトピーマウスにおけるアレルギー病態制御の分子機構

Research Abstract

NC/NgaマウスのIgE標的抗原としてダニアレルゲンを想定して検索した結果、173kDa抗原が昨年度検出された。本抗原の分子量が高分子ダニアレルゲンDer f 14/M-177のそれと酷似していることに鑑み、同分子のcDNAクローニングを試みた。Der f 14 cDNAは1666アミノ酸からなるポリペプチドをコードしており、apolipopholin, vitellogenin, apolipoprotein B-100といった脂質輸送タンパク質と相同性を示し、更に最近同定されたダニアレルゲンEur m 14とも約90%の相同性を有していた。同抗原のヤケヒョウヒダニホモログ(Der p 14)のC末端断片が強力なTh2応答を惹起しうることから、本分子のNC/Ngaマウスにおける高IgE血症誘発抗原としての重要性が推察された。今後、本分子の組換えタンパク質発現系を構築してIgE・T細胞エピトープの検索を進めることで、allergen-definedなアトピー発症モデルの構築に繋がることが期待される。
昨年度、NC/Ngaマウスの病態進行に伴いリンパ組織の腫脹が認められることを見いだしたが、cell population解析から主にB細胞の割合が増加していることがわかった。更に、同B細胞におけるIL-4 receptor感受性が高まっていること、血中IL-18レベルが発症とともに著明に上昇することが明らかとなった。これらの観察から、1)B細胞の過増殖、2)過剰のIL-4シグナル、3)IL-18依存性のinnate typeのIgE誘導機構、が複合して同マウスの高IgE産生に関与している可能性が示唆された。
