Research Abstract

Polo-like kinase(Plk)は、胎盤に強く発現が認められた非常にユニークな遺伝子である[Hamanaka et al. Cell Growth & Differ.5:249-257,1994]。Plkは、蛍光抗体法により細胞分裂周期のS期の細胞質と核内に発現が認められ、またアンチセンスPlkのマイクロインジェクション実験では細胞分裂時に現われる微小管の形成に必須の遺伝子である事が明らかにされた[Hamanaka et al. J. Biol. Chem.,270:21086-21091,1995.]ことより、細胞分裂の盛んな細胞でPlkは発現が増強していることが予想される。われわれは、マウスの胎盤,胎児,卵巣における実験で、細胞分裂の盛んなtrophoblastや胎児臓器などでPlkの強い発現を確認し、さらに胎盤や黄体の分化の過程にもPlkが重要な役割を果たしている事を報告し[Takai et al. Reprod. Fertil.Dev.,11:31-35,1999]、またヒト正常子宮内膜では増殖期後期から分泌期前期の腺管や間質でPlkが強く発現していることも報告した[Takai et al. Reprod. Fertil.Dev.,12:59-67,2000]。しかし、細胞分裂が盛んな癌細胞では、Hela細胞などの培養癌細胞でPlk mRNAの発現が認められるという事が報告されている[Hamanaka et al. Cell Growth & Differ 5:249-257,1994.]だけで、plkは臨床的にどのような癌細胞で,どのような分化度の細胞で,どの程度の発現が認められるのかなど全く解析されていない。そこで、婦人科癌(子宮頚癌、子宮体癌、卵巣癌)の増殖機構や分化機構と、細胞分裂期に特異的に発現するPlkとの関連について検討し、さらにその癌に罹患した患者の予後を検討する事とした。今年度は、卵巣癌[Takai et al. Cancer Lett.,164:41-49,2001]と子宮体癌[Takai et al. Cancer Lett.,169:41-49,2001]でのPlkの発現を検討し、分化度の低い症例や臨床進行期の進んだ症例に有意に多く発現していることを明らかにした。
