Research Abstract

本研究は「L-band ESRを用いて口腔内領域で産生されたNOをin vivoリアルタイム計測し,その分布領域をin vivo ESR-CT imaging systemを用いて3次元画像化すること」を最終目標とし,平成12年度の実績を基盤として平成13年度では,Fe-DTCS錯体によるNOトラツプ手法をL-band in vivo ESRに移行させ,病態モデルを用いたin vivo NOトラッピンクおよび顎顔面部におけるNO分布領域の画像化に着手するため,(1)LPS処置Wistar rat(LPS腹腔内投与により誘導されるi-NOSは,生体内でNOを産生する)血液中NOのX-band in vivo ESRを用いたFe-DTCS錯体による捕捉・検出,(2)LPS処置ICR mouseのL-band in vivo ESR測定の実験プロトコルを計画した。LPS投与6時間後,Fe-DTCS錯体を尾静脈より投与し,採取した血液サンプル内にNOの存在を実証するNO-Fe-DTCS錯体のESRシグナルを検出することができた。この実験結果を基礎データとして,LPS処置ICR mouseのL-band in vivo ESR測定に移行した。L-band in vivo ESR測定ではマウス腹部でのESRシグナル検出が限度であり,頭部領域での検出およびその画像化までは今のところ達成していない。原因として誘電損失による感度低下,装置の検出限界と生体内NO錯体濃度との差,検出される広いシクナル線幅など多くの問題点が挙げられる。現在これら問題点を克服のため,ハード・ソフト両面からの改良を行っており,標記課題達成のための実険を遂行中である。
