Research Abstract

【目的】SPARC/osteonectinは,分子量43-kDaのmatricellular proteinである.私達はこれまでに腫瘍の悪性化とSPARCの役割について検討してきたところ,SPARCが腎癌細胞の運動性を亢進すること,マウスB16メラノーマ細胞での転移性とSPARC合成が相関すること,VEGFが血管内皮細胞からのSPARC合成を誘導することなどを報告してきた.本助成金により平成13年度は,SPARCノックアウトマウスに形成された腫瘍および,SPARCノックアウトマウス由来線維芽細胞の形質転換細胞の性状を解析し,癌の発生と進展に関して検討した.【結果と考察】インビトロでのDMBA-TPAを用いる形質転換実験では,SPARCノックアウトマウス由来線維芽細胞で形質転換したコロニー形成が高率に観察された.化学発がん(DMBA-TPA)による腫瘍の形成は,SPARCノックアウトマウスにおいて早期に出現したが,SPARCノックアウトマウスにおいて形成された腫瘍は良性の乳頭腫であったのに対してワイルドタイプでは組織学的に悪性化を示していた.SPARCノックアウトマウス由来線維芽細胞では,DMBAによるDNA adduct形成量が多く,この理由はDMBA代謝能や初期の結合能の差に基づくものではなく,DNAの修復能の低下によるものと考えられた.このようにSPARCは,腫瘍の発生と進展において2面性の作用を持つことが示唆された.
