Research Abstract

骨髄微小環境内におけるB細胞産生にいかなる細胞表面分子が関与しているかについて、我々はいろいろな方面から研究を進めているが、平成12年度の研究期間に新たな知見を得たので報告する。
1.本研究計画にも記述していたように、我々はLong-term bone marrow cultureにおけるB細胞産生を阻止する新しいモノクロナール抗体を樹立した。今回、我々はこの抗体の認識するB細胞表面分子を同定する事に成功した。まず抗体の認識する膜タンパクを生成し、そのタンパク質のN末端アミノ酸配列を決定した。コンピューターサーチの結果、それが接着分子の一つであるICAM-2(CD102)分子である事が判明した。この事を裏付けるために遺伝子導入実験を施行したが、確かに我々の抗体が特異的にICAM-2分子を認識する事を確認したさらにこの抗体を正常マウス(Balb/c)に投与すると、骨髄における成熟B細胞産生が著明に減少する事が判明した。つまりin vitroのみならずin vivoにおいても同様の結果が得られた事になる。現在これらの知見をまとめ、論文投稿準備中である。
2.一方で、B細胞表面分子であるCD72に注目し、その機能解析を進めている。この分子からのシグナルが活性化を伴い成熟B細胞のサバイバルを誘導する事を以前、我々は明らかにした。今回、この分子の発現あるいはシグナル伝達に別の細胞表面分子であるFcレセプターが重要な役目を果たしている事を確認した。FcレセプターはB細胞におけるシグナル伝達分子でもあり二つの分子がクロストークして機能調節を担っているのかもしれない。このような報告は世界的にみても未だされておらず現在、これらの知見を確かなものにするためにさらなる解析を進行中である。
