Research Abstract

1.DNAポリメラーゼβ・8kDaドメインの大量発現
DNAポリメラーゼβ(polβ)・8kDaドメイン遺伝子のN末側にHis-タグを挿入して、その遺伝子を大腸菌へ組み込んだ。そして大腸菌を培養して遺伝子を発現させて、ニッケルカラムで簡単にかつ大量に8kDaドメインを供給することに成功した。培地に^<15>NH_4Clを添加して蛋白質を安定同位体ラベルすることにより、2次元NMR測定用の8kDaドメインサンプルを提供できるようにした。
2.DNAポリメラーゼβ・8kDaドメインと阻害剤の2次元HMQC-NMR解析
長鎖脂肪酸のリノール酸およびネルボン酸、胆汁酸ステロイドであるリトコール酸がpolβ・8kDaドメインの一本鎖DNA結合活性を阻害したので、阻害剤の8kDaドメインへの結合を^1H-^<15>N HMQC-NMRで解析した。長鎖脂肪酸は8kDaドメインのLeu11,Lys35,His51,Thr79と、リトコール酸はLys60,Leu77,Thr79と結合することを見いだした。長鎖脂肪酸・リトコール酸ともにカルボキシル基を有しており、カルボキシル基がLys残基と水素結合することが考察された。
3.糖脂質(Sulfo-quinovosyl-monoacyl-glycerol, SQMG)の化学合成
長鎖脂肪酸よりも10倍以上強くpolβを阻害する糖脂質(SQMG)の有機化学合成に成功した。また各種類似体・誘導体合成にも成功して、阻害剤の構造活性相関を明らかにした。SQMGと8kDaドメインの結合のNMR解析を現在実施している。
4.DNAポリメラーゼβ・8kDaドメインの変異蛋白質の作製
遺伝子工学的手法を用いて、長鎖脂肪酸およびリトコール酸が結合する上述の8kDaドメインのアミノ酸残基をAlaに変異させた蛋白質の作製に成功した。阻害剤の変異蛋白質への結合について、野生型(非変異型)との比較はこれから実施するところである。
