Research Abstract

(1)解離した細胞(生殖細胞およびセルトリ細胞)からRNAを抽出し、これを鋳型としてsingle cell RT-PCRをおこなった。コントロールとしてbeta-tubulinの増幅をおこない良好な結果が得られたことから、single cell PCRのシステムは確立したと考えられる。しかしながら発現を調べたいBMP、BMP受容体遺伝子の増幅は、遺伝子発現が微量であるため現在のところ成功しておらず、さらにPCRの条件を検討していく必要がある。
(2)BMPによってアポトーシスが誘導されることから、プロラクチンによって誘導されたBMPが直接精原細胞に働くと考えられた。しかしながら、BMPは精原細胞期に一定に発現量を保つことから、BMPの活性を阻害するアンタゴニストが調節因子として働いている可能性が考えられ、それらのクローニングと発現解析をおこなった。イモリ初期胚から既知のBMPアンタゴニストのnoggin、chordin、follistatin、cerberus、DAN、gremlin cDNAをそれぞれクローニングし、イモリ精巣での発現を調べたところDANおよびchordinの発現が比較的高いことがわかった。さらに、器官培養においてプロラクチンとFSHで処理したときのDANとchordinの発現をRT-PCRによりそれぞれ調べたところ、発現の変化はみられなかった。これらの結果から、他のアンタゴニストが関与する可能性とタンパク質レベルでBMPおよびアンタゴニストの発現が変化する可能性が考えられた。
