Research Abstract

DNAマイクロアレイを用いたてんかん関連分子の検索と候補遺伝子の絞り込み
野生型マウス脳とグルタミン酸トランスポーター(GLT-1)ノックアウト(KO)マウス脳からそれぞれmRNAを抽出し、それらを鋳型にしてランダムプライマーを用いてcDNAプローブを作製し、DNAマイクロアレイに適用した。野生型マウスとGLT-1 KOマウスの遺伝子発現のプロフィールの差異をコンピューター解析し、GLT-1 KOマウスにおいて特異的に発現が増加あるいは減少している遺伝子を検索し、数多くの候補遺伝子が見いだされた。その多くは、従来遺伝子機能の不明なESTタグがほとんであったが、昨今、マウスの全長cDNA配列の決定が進み、それらESTタグの配列と報告されているマウスの全長cDNA配列を比較検討することにより、その機能がある程度予測可能でかつ、脳に発現していることが確認できる候補遺伝子が見いだされた。
それら候補遺伝子には、様々なシグナル伝達に寄与すると考えられるセリン・スレオニンリン酸化酵素、ユピキチン連結酵素に相同な構造を持つもの、カチオンチャネル、カルシウム輸送酵素、低分子量GTP結合蛋白質等の機能を持つ興味あるものが含まれている。
現在、これらの候補遺伝子に特に注目し、1)ヘテロ型GLT-1 KOマウスにおいても、発現に差異がでるものを選出する、2)電気ショックなどで人為的に痙攣発作を誘発したマウスと正常マウスを用いてDNAマイクロアレイを行い、発現に差異が出た遺伝子と1)で発現に差異が出た遺伝子を比較し、一致するものは、てんかん発作自身によって発現が変化した遺伝子と判断し、本研究の目的遺伝子から除外する、などの検討を行い、てんかんの発症や予防に関連し、さらに、新しい抗てんかん薬の標的になりうる分子の絞り込みを行っている。
