Research Abstract

マウス,ラットを用いて、骨髄内(Intra-bone marrow : IBM)に骨髄細胞を移植(IBM-BMT)すると、従来の方法(静脈内[IV]に骨髄細胞を移植するIV-BMTや門脈[PV]内に骨髄細胞を移植するPV-BMT)よりも、放射線量を減量しても生着率が高く、造血の回復も速やかであることを発見した(Blood 97:3292-3299,2001)。それ故、ヒトへの応用を視野に入れて、モンキーの実験を開始し、次のような結果が得られた。
i)末梢血の混入を防ぐため、従来の腸骨を用いたAspiration Method (AM)に代わって、長管骨を用いたPerfusion Methodを開発(Stem Cells 18:453-456,2000)し、長管骨と腸骨での骨髄細胞の採取率と年齢的変化を検討し、モンキーでは安全性の上からも上腕骨が最適であることを明らかにした(Stem Cells In press)。
ii)灌流法に用いる骨髄採取セットを考案中で、骨片が骨髄内に混入せず、しかも摩擦熱の少ない電動式の骨髄採取セットを開発中である(特許申請中)。
iii)"Conditioning"としては、マウスの実験でDonor lymphocyte infusion(DLI)を用いれば、放射線量を減量しても生着不全が起らないことを見出した。
