Research Abstract

昨年度は発癌抑制作用を有するlycopeneおよびlactoferrinをそれぞれ産生する大腸菌およびBacteroides uniformisを作製し腸管に定着させることで、azoxymethane(AOM)で誘発される大腸前癌病変aberrant crypt foci(ACF)の形成が抑制されることを示した。本年度はラクトフェリン産生腸内菌であるBacteroides uniformis TCTK101株を用いて以下の点を明らかにした。
1.ヒト由来ラクトフェリン遺伝子を大腸菌-BacteroidesのシャトルベクターpVAL-1に導入して得られたプラスミドpVLFKを用いてBacteroides uniformis BU1001株を形質転換することでラクトフェリン産生B. uniformis TCTK101株を作製した。900bpのラクトフェリンDNAプローブを用いたノーザンハイブリダイザーションによって2.2kbのラクトフェリンmRNAが合成されていることを確認した。
2.B. uniformis TCTK101株においてラクトフェリンタンパク質は外膜に局在していた。
3.B. uniformis TCTK101株の一夜培養液をF344ラットに飲料水として摂取させたところ、ベクター対照株であるB. uniformis TCTK11株投与群と比べて脾臓単核細胞のYAC-1細胞に対する細胞障害活性が有意に上昇した。
