Research Abstract

我々はチトクロームP450 (CYP)の1分子種であるCYP2A6が4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK)などのたばこ煙中のがん原性N-ニトロソアミン類を代謝的に活性化すること,および喫煙歴を有する肺がん患者においてCYP2A6の遺伝子多型である全欠損型変異を有するヒトの頻度が対照と比較して有意に低いことを明らかにした.これらの事実から,CYP2A6がたばこ煙中のN-ニトロソアミン類を代謝的活性化し,肺がんを誘発する可能性が示唆された.したがって,CYP2A6が上述のN-ニトロソアミン類を代謝的に活性化するか否かをin vivoで検討することは非常に重要である.そこで本研究ではヒトCYP2A6 cDNAを導入したトランスジェニックマウスを樹立し,CYP2A6がたばこ煙中のN-ニトロソアミン類を代謝的活性化し,発がんに寄与しているか否かをin vivoで解明することを目的とした.CYP2A6の開始コドンからpoly(A)付加シグナルまでを含むCYP2A6 cDNAの全長を単離し,プロモーター,エンハンサーおよびインスレーターが導入された発現プラスミドを構築した.続いて,断片化した発現プラスミドを受精卵へ注入した後,偽妊娠マウスへ移植した.誕生したマウスの尾からゲノムDNAを抽出し,PCR法にて導入遺伝子を検出した.このようにしてF0世代を樹立した.F0マウスを通常マウスと交配することにより,F1世代を得た.現在,導入遺伝子が検出されたF1世代マウスを用いて,導入遺伝子コピー数の揃った個体同士を交配させ,F2世代を作成中である.導入CYP2A6遺伝子をホモで持つ動物F2マウスが計画どおりに作出されれば,本研究の究極的な目標により一層近づくことができると考えられる.このヒト型Tgマウスを用いると,発癌性試験におけるヒトの酵素の役割や,作出したヒト型マウスを他のin vivoでの遺伝子変異検出系マウスとの交配などによって,このCYP2A6研究領域のさらなる進展が期待される.
