染色体脱凝縮により活性する遺伝子群のゲノム局在様式と発現のメカニズム

Research Abstract

1.マクロアレイフィルターを用いた発現プロフィルの解析により,ラット小脳顆粒神経細胞の分化過程で発現が誘導される54個の遺伝子を同定した.このうち,いくつかの遺伝子の特定領域クロマチンの構造変化をDNase I/定量PCR法を用いて解析したところ,トポIIβの活性に依存した発現誘導を示す遺伝子群(Class I遺伝子)では培養経時的にDNase I感受性が上昇し,トポII特異的阻害剤ICRF-193で活性を阻害した場合その上昇は抑制された.しかし,発現誘導が阻害剤に感受性を示さない(その誘導にトポIIβの活性を必要としない)遺伝子群(Class II遺伝子)の場合,培養開始直後から感受性は高く,以後大きな変化は認められなかった.これらの結果は,凝縮状態にあるClass I遺伝子クロマチンをトポIIβが脱凝縮することにより,転写誘導が可能になることを示唆している.
2.次に,これらの遺伝子が局在するアイソコア構造をヒトのゲノム情報に基づいて解析した.その結果,Class I遺伝子のうち,amphiphysin Iやphosphodiesterase 4B遺伝子などは高度に凝縮したクロマチン構造をとる,AT含量の高いアイソコア(L2)に存在したが,Sodium channel β1やRAD GTPase遺伝子などはむしろGCに富むアイソコア(H2)に局在した.すなわち,トポIIβに依存した遺伝子発現の誘導は,アイソコア構造のみに依存するのではなく,より局所的なクロマチンの凝縮状態に支配されることが明らかになった.
