Outline of Annual Research Achievements

本年度は、昨年に引き続きミナミトビハゼの生体リズムの形成機構の解明に取り組んだ。また、本種の生息環境・塩分濃度は潮汐に応じて大きく変化していることから、環境浸透圧の変化が潮汐情報因子の１つとして働いている可能性が考えられた。そこで、本種の浸透圧調節機構についても調べた。
１生体リズムの解明
本種の時計遺伝子（Period1（Per1）、Per2、Cryptochrome2（Cry2）およびCry3）は脳および網膜において日周リズムを示した。恒暗条件下におくと、脳において調べた全ての時計遺伝子の発現量が明暗条件下に比べ大幅に下がり、一方、網膜においてはPer1およびPer2の発現量が明暗条件下に比べ減少した。また脳ではPer1およびCry2のみが概日変動を示した。一方、網膜において調べた全ての時計遺伝子の発現が概日リズムを示した。これらの結果から、本種において網膜が概日時計のペースメーカーの器官の１つであることが示唆された。
２浸透圧調節機構の解明
本種を淡水または海水へ移行したときの血漿ナトリウムイオン濃度を調べたところ、約160 mM前後でほぼ一定で変化が見られなかった。鰓と皮膚においてNa+/K+ATPaseα1の発現は淡水で上昇し、一方、Na+/K+ATPaseα3の発現は海水で上がった。さらに脳-下垂体ではプロラクチン（PRL）の発現は淡水で高く、一方イソトシン（IT）の発現は海水で高くなった。これらの結果から、PRLおよびITが環境浸透圧に応じて鰓や皮膚のNa+/K+ATPaseの働きを調節することで、血漿ナトリウムイオン濃度を一定に保てることが示唆された。以上より、本種の潮汐性生体リズムの形成には時計遺伝子は関わっておらず、浸透圧調節ホルモンなどが関わっている可能性が示唆された。
