Outline of Annual Research Achievements

本研究は、「人類のアポクリン腺関連形質決定因子: ABCC11のin vivoにおける生理的機能解析を軸に、アポクリン腺分泌機構の制御を担う分子システムを統合的に解明することを目的とし、アポクリン腺関連形質疾患: 腋臭症の治療薬探索に応用することを目指す研究」である。平成26年度は、主に以下の項目に取り組んだ。

●ABCC11発現マウスの解析：　平成25年度までの研究で得られたABCC11発現マウスの解析を進めた。ABCC11タンパク質の組織内局在を検証するために、ABCC11発現マウスから摘出した各種組織の凍結切片を用いて、免疫組織化学法による検討を行った。その結果、特に乳腺、凝固線の管腔側頂端膜においてABCC11の発現が確認された。これまでの結果と合わせて考えると、ABCC11がapical側に局在することが確かめられた。野生型マウスとABCC11発現マウスとにおけるメタボローム差異解析の結果、両者の間で変動があるピークを見出すことができた。ただし、構造同定には至らなかった。候補物質の標品入手について、有機合成を視野に入れながら、今後も検討を続けていく予定である。

●ABCC11阻害剤の探索：　ABCC11発現膜ベシクルを用いて、[3H] dehydroepiandrosterone-sulfate（DHEAS）を基質とする輸送活性測定系を構築した。反応時間を10分間に定め、試験化合物（最終濃度10 μM）の存在がABCC11によるDHEAS輸送に与える影響を評価することで、試験化合物による阻害率を求めた。現在、輸送活性を低下させる物質を探索している。
