骨格筋-脳-心連関を見据えた心不全におけるBDNFの役割の解明

Research Abstract

マウスにおける運動能力および骨格筋ミトコンドリア機能におよぼす脳由来神経栄養因子(BDNF)-TrkBシグナルの役割を検討した. その結果, BDNFもしくはTrkB受容体アゴニストの投与群は対照群に比較し, 運動能力および骨格筋ミトコンドリア機能を有意に増加した. その機序として, BDNFもしくはTrkB受容体アゴニストの投与は骨格筋におけるAMPK-Sirt1-PGC1α経路を活性化することを明らかにした. したがって, BDNF-TrkBシグナルの活性化は運動能力を制御する重要な役割を果たす可能性を示唆した.
BDNFノックアウトマウスにおける運動能力および骨格筋ミトコンドリア機能を検討した. BDNFノックアウトマウスは野生型マウスに比較し, 運動能力および骨格筋ミトコンドリア機能が有意に低下した. したがって, BDNFは運動能力および骨格筋ミトコンドリア機能を直接的に制御する可能性が示唆された. これらの結果により, BDNF-TrkBシグナルの活性化は, 骨格筋ミトコンドリア機能を調節することで, 運動能力の調節に中心的な役割を担っていることが示唆された.
本研究によって、運動耐容能におけるBDNFの役割が明らかとなれば、BDNFは精神機能のみならず、運動能力を反映するを示す新しい指標となりうると考える。また、動物研究ではBDNF投与によって、精神機能の改善が報告されている、一方で、糖・脂質代謝を改善することから様々な慢性疾患モデルの運動能力を改善させる可能性がある。従って、老化、糖尿病や癌などの慢性疾患に対する新しい機序の治療薬となり得ると考えられ、臨床応用を見据えた研究へと発展できると考える。したがって、本研究の結果は幅広い疾患に応用できる可能性があり、健康社会の実現に寄与することが期待される。
