Outline of Annual Research Achievements

哺乳類中心体非依存的スピンドル形成の重要因子のひとつとして、オーグミン複合体を中心に研究を行っている。昨年度に引き続き、複合体のサブユニットの中でもコアと考えられるAug6に着目し解析を行った。樹立したAug6全身性欠損ヘテロマウスを用いて交配を行い、ホモ欠損マウスの作出を行ったが、ホモ欠損マウスは発生初期に耐性致死となることが判明した。欠損胚では、野生型に比べ、胚盤胞期の発生が遅延し、細胞分裂の停滞及び失敗を示す凝縮染色体や異常形態の核が観察された。
また、卵子および初期胚の細胞分裂解析系構築のため、GFP-tubulin発現マウス及びスピニングディスク共焦点顕微鏡による3次元イメージング及びエレクトロポレーションによる遺伝子発現システムの導入を行った。それにより、初期胚の細胞分裂時には、複数の微小管形成中心が現れ、それらが２つの極に集極するような特徴的な形式で細胞分裂が遂行されることが観察された。そこで、交配したAug6欠損胚で同様に観察したところ、微小管形成中心がスピンドル内部で散乱し、それに伴い分裂の遅延が起こることが明らかになった。中心体マーカーの解析により、この時期の胚は、成熟した中心体が形成される前であることが判明した。これらのことから、オーグミンが中心体形成前の初期胚の細胞分裂に重要な役割を持つことが示唆された。
上述のような、微小管形成中心の集極化による二極化紡錘体形成は、癌細胞等の中心体が増幅した細胞でも観察されることから、中心体増幅の細胞でも検証を行ったところ、オーグミン経路を阻害する処理によっては、中心体の集極化に障害が観察された。これらの結果により、オーグミン経路が初期発生のみならず癌細胞の細胞分裂でも共通する重要な役割をもつことが示唆されており、現在これらを成果としてとりまとめ論文作成にとりかかっている。
