頭足類単眼胚が浮き彫りにする眼の発生メカニズムの共通性と多様性

Outline of Annual Research Achievements

ヒメイカにおいて正中に眼が1個しかできないCyclops(単眼)胚が希に現れることから、その初期発生において単一の眼原基が左右に分割して2つの眼が形成されることが示唆されている。本研究において、ヒメイカ初期胚を用いた薬剤阻害実験系を確立し、1-Azakenpaurone処理による阻害実験からWntシグナリングの阻害によって単眼胚が誘導されることを見いだした。1-Azakenpaurone処理によって誘導した単眼胚サンプルを用いて、ファロイジンおよび抗アセチル化チューブリン染色によって分化した神経系を可視化し、単眼胚においては神経細胞が分化しているものの、正常胚に見られるような左右相称の脳神経節、視葉が形成されないことを見出した。また、1-Azakenpaurone処理の臨界期に当たるヒメイカ胚盤期中期において、脊椎動物の眼原基形成に関与している転写因子群のうちPax6、Otx1/2、Six3/6がの眼原基で発現していることを確認し、Wntシグナリングによってこれらの転写因子の発現・および眼原基の位置が制御されていることが示唆された。上記の結果をまとめて原著論文を作成した。また、海外渡航制度において、フロリダ大における軟体動物の比較ゲノム研究に従事し、眼原基形成に関与している転写因子群の相同遺伝子を同定し、その調節領域についての研究を遂行した。
