Research Abstract

ペルフルオロアルキルカルボン酸(Perfluoroalkyl carboxylates ; PFCAs)は多数の炭素鎖の異なる同族体をもつ残留性有機汚染物質である。PFCAsの鎖長とその毒性は密接な関連があるとされることから、これらの体内動態のデータはPFCAsの毒性の把握において重要な情報となるとされる。申請者は長鎖を含むさまざまな鎖長のPFCAsについてマウス投与実験による詳細な体内動態(吸収・分布・代謝・排泄)の検討と血中濃度モデル化を行った。PFCAsの鎖長とその毒性は密接な関連があるとされることから、これらの体内動態のデータはPFCAsの毒性の把握において重要な情報となる。また本研究では母乳への移行についても独自に作成したマウス搾乳機を用いて検討を行った。本研究で得られた知見は以下のとおりである。(1)マウスを用いた動物実験によると短鎖PFCAsは急速に血中から尿に排泄される一方、長鎖PFCAsは肝臓に蓄積され、血中濃度半減期が長い。総クリアランスは炭素鎖9,10,11付近で最も低く、残留性が最も高くなる。(2)母乳中への移行係数は中長鎖PFCAs (C8-C12)の間には一定であるが、それ以上の鎖長では分配率が上がる。母乳を通じた乳児への曝露については鎖長13以上のPFCAsではさらに警戒が必要な可能性を示す。(3)ヒトにおいて、尿と胆汁へのクリアランスを評価し、排泄能力の違いはあるものの、マウスと共通の傾向を見出した。(4)さらにマウスの脳中とヒトの脳脊髄液への移行を比較したところ、マウスにおいては鎖長が長くなるにつれ、血清と比較した脳中のPFCAs割合が増加していた。しかしながらヒトの脳脊椎液ではその傾向はみられず、PFCAsはヒトの血液脳関門は透過しないと思われる。

Reason

本年度、環境汚染物質の体内動態を明らかにするために、動物実験で得られたデータの解析に精力的に取り組んだ。また合わせてヒト生体試料(血清・尿・脳脊椎液等)の分析にも取り組み、動物実験で得られた結果のヒトへの外挿の検討を行った。現在論文を投稿中であり、おおむね期待通りの進展をしたと考えられる。
