Outline of Annual Research Achievements

ミジンコ類は環境依存型の性決定様式を有しており、単為生殖によって好適環境ではメスを、生存に不向きな環境下ではオスを産出する。オス産出を引き起こす環境要因は日照時間や水温、栄養状態など数多く報告されているが、遺伝的背景の異なる系統間でも各環境シグナルへの応答能が異なるため、実験室環境で安定して雌雄の産み分けを誘導できる実験系は確立されていなかった。私はこれまでに、海外で系統化されたミジンコ(Daphnia pulex)のWTN6系統が日長条件に応じて雌雄を産み分けできることを発見した。昨年度の研究でミジンコのオス誘導に内因性の幼若ホルモンが関与している可能性が高いことを示すデータを多数取得した。
本年度は更に詳細な解析を進めるために以下の研究をおこなった。性決定期の幼若ホルモンシグナルの制御に関与している因子を探索するために、長日（メス誘導）条件と短日（オス誘導）条件で飼育したWTN6系統の母親個体のトランスクリプトーム解析をおこなった。その結果、短日条件で統計的有意に発現変動している遺伝子としてイオンチャネル型グルタミン酸受容体に関連する遺伝子を見出した。イオンチャネル型グルタミン酸受容体のサブタイプ特異的な活性化剤や阻害剤の投与によるオス誘導に与える影響を調べたところ、N-methyl-D-aspartate (NMDA)型グルタミン酸受容体がミジンコのオス誘導を引き起こす幼若ホルモンシグナルの上流制御因子として作用している可能性が高いことが明らかになった。現在はトランスクリプトームから得られた他の候補遺伝子についても個別に機能解析を進めている。
