Outline of Annual Research Achievements

平成25年度に見出したペプチダーゼ耐性を有するNK3受容体アゴニストのin vivoにおける生物活性評価を、卵巣摘出ヤギのMUAボレーを指標として共同研究により実施した。その結果、本ペプチドが既知NK3受容体選択的アゴニストであるsenktideに比べて高い持続性を示すことを見出した。本検討により見出された新規NK3受容体アゴニストペプチドは、GnRHパルス発生機構の解明に向けた新たなツールとしての利用が期待される。
続いて、平成25年度に新たに見出した新規高活性NK3受容体選択的アゴニストについて、生体試料中における分解特性を解析した。その結果、senktideのN末端アミノ酸をD-グルタミン酸に置換することで、ペプチダーゼ耐性が向上することを見出した。一方、本ペプチドのin vivoにおける作用時間はsenktideと同等にとどまったことから、ペプチドのペプチダーゼ耐性はin vivoにおける作用持続時間と直接相関しないことが示唆された。本検討結果は、長時間作用型NK3受容体アゴニストの創製に向けて重要な知見を与えるものと考えられる。
また、生体内でのGPR54の局在解析、及び、キスペプチン類の標的探索を目的として、ラットキスペプチン(rKp-14)の配列に基づき、4種類の光アフィニティープローブを設計・合成した。合成したプローブはいずれもGPR54に対するアゴニスト活性を維持した。一方、GPR54発現細胞より調製した膜画分に対して光親和性ラベル化実験を実施したところ、受容体の標識には高濃度のプローブを要することが判明したことから、標識における特異性と結合親和性の向上が今後必要であることが示唆された。
