植物の低温馴化過程におけるGPIアンカータンパク質の機能と凍結耐性との関連

Outline of Annual Research Achievements

低温馴化過程での細胞膜組成の変化は、低温環境下での植物の生存を左右すると考えられており、本研究では細胞膜脂質修飾型タンパク質の一種であるGPIアンカー型タンパク質(GPIAP)の低温馴化における機能特性を解析した。昨年度までは、GPIAP濃縮画分を初めとした様々な画分のショットガンプロテオーム解析により、163種類ものGPIAPの低温馴化応答性を明らかにすることができ、その中からAt3g04010を選出して発現パターンや凍結耐性への関連性を明らかにした。本年度はまず、TTC assayにより、At3g04010の凍結耐性への影響を組織的に評価した。その結果、At3g04010欠損変異体(at3g04010)は凍結融解後の回復過程で、茎頂の再生長の著しい遅延もしくは停止を引き起こしていることが分かった。さらに、このAt3g04010は、ドメイン構造などからβ-1,3-glucanaseの一つであると考えられるため、β-1,3-glucanaseの基質であり、細胞壁成分の一つでもあるカロースの観察と定量を行った。その結果、師部組織のカロース量は、野生型株では低温馴化7日目にかけて減少し、at3g04010においてはその減少が見られないことが分かった。さらに、蛍光タンパク質mCherryでAt3g04010を標識して顕微鏡観察したところ、師部組織の細胞間隙でパッチ状に分布することが明らかになった。このような組織内分布パターンはカロースの分布とも酷似するものである。さらに、これらの蛍光強度は低温馴化7日目にかけて2.7倍まで高くなっていくことが分かった。カロースは細胞間コミュニケーションや師部組織の通導性に影響を及ぼすことから、At3g04010は低温馴化過程でカロースの分解を通じて組織の師部輸送能を上げ、凍結融解後の茎頂の再生長を促す役割があるものと考えられる。
