Outline of Annual Research Achievements

F1-ATPaseは、ATP(生体内のエネルギー分子)の合成・分解を担っている。この酵素は、軸回転という構造変化を通して、その機能発現に至る。申請者は、この回転型分子モーターについて、その動作機構の解明に取り組んできた。一連のF1-ATPaseの回転は、基質であるATPのイベント(ATP結合、生成物解離)によって引き起こされる。F1-ATPaseの軸回転の全貌を知るには、これらの過程を個別に解明する必要がある。研究手法としては、蛋白質の構造変化を原子レベルで追跡でき、その構造変化を自由エネルギーという変化量で定量的に測れる分子動力学計算を用いた。
これまでの申請者の研究により、両方の(ATP結合、生成物解離によって引き起こされる)回転ステップのメカニズムが明らかになった。どちらの構造変化においても、構造変化が自発的におこる発熱的な自由エネルギー地形であった。さらに両ステップとも、ATPaseファミリーに普遍的に存在するWaklerモチーフというアミノ酸のグループが、全体の大きな構造変化を引き起こすトリガーの役割を果たしていることも分かった。このATPaseに共通のアミノ酸モチーフが構造変化に重要な役割を果たしているという結果は、他のATPaseファミリーにおいても、同様な構造変化のメカニズムを持つことを示唆する。また、このシミュレーションの過程では、既存の実験結果とも整合する精度で、未だ構造が明らかになっていないstateについても予測できた。以上の結果は、多くの学術論文や、総説、学会などの機会で発表させて頂いた。
