Outline of Annual Research Achievements

本研究の目的は、腎エリスロポイエチン産生細胞(以下、REP細胞)の形質転換で生じた筋線維芽細胞による腎筋線維芽細胞全体に対する寄与度および、その形質転換の分子機序を解明することで、腎性貧血の発症と腎線維化の進行を抑制する新規治療法『REP細胞再生療法』開発の基盤を作製することである。本研究課題にて作成した腎性貧血モデルマウス(Inherited Super-anemic Mice, ISAM)より単離したREP細胞の網羅的トランスクリプトーム解析を行った。本年度には、マイクロアレイ解析に追加して、RNAシークエンス解析も行った。マイクロアレイ解析により、内皮細胞維持に必要な増殖因子のリガンドの発現が形質転換とともに低下していることが明らかになった。そこで、腎臓病時における内皮細胞維持機構の破綻の解析を、ノースウエスタン大学腎臓内科との共同研究にて行った。内皮細胞維持の分子メカニズムを明らかにするために、血管内皮に発現する受容体チロシンキナーゼの自己リン酸化部位を質量分析にて同定した。さらに、病態モデルでのリン酸化、シグナル伝達の変化の解析を行った。また、同シグナル増強薬を入手し、糖尿病性腎症マウスモデルにおいて、その線維化に及ぼす効果を検討し、今後につながる予備的結果を得た。
