原口から脊索を生み出した進化過程のＣｈＩＰ－ｓｅｑ法を用いた解析

Outline of Annual Research Achievements

１）サンゴ胚に薬剤処理を施し、Wnt/β-cateninシグナルを活性化したところ、brachyuryの発現が活性化され、外胚葉全体に発現が広がった。一方、Wnt/β-cateninシグナルを抑制したところ、brachyuryの発現が弱まるもしくは消失した。さらに、サンゴbrachyuryの上流約1 kbをクローニングし、Xenopus胚を用いてLuciferase reporter assayを行った結果、サンゴTcfによってレポーターの発現が制御されることが示された。
２）Xenopus特異的な二つのbrachyuryパラログ遺伝子brachyury（主に尾芽に発現）およびbrachyury2（主に脊索に発現）のエンハンサー候補配列の活性について、トランスジェニックレポーター解析を行った。その結果、brachyury U1領域は脊索とともに体節でもエンハンサー活性を持つが、U2領域と組み合わせることで体節でのレポーター遺伝子の発現は弱まり、脊索での発現がより顕著になることが示された。一方、brachyury2のU1領域は体節でのエンハンサー活性が弱く、脊索でのエンハンサー活性が強かった。このU1領域の活性の違いが、二つの遺伝子機能の分化に寄与した可能性が高い。さらに、各brachyury機能阻害胚のRNA-seq解析を行った結果、Brachyury2特異的な標的遺伝子候補として、脊索細胞の液胞化や細胞形態に関わる遺伝子が多く見つかった。
３）サンゴbrachyuryの機能阻害胚を用いたRNA-seq解析の結果同定された標的遺伝子候補のうち、brachyury機能阻害胚で発現の減少した遺伝子は原腸胚期で主に外胚葉で発現しており、逆に機能阻害胚で発現が増加した遺伝子は内胚葉で発現することが示された。これは、サンゴBrachyuryが原口において外胚葉と内胚葉の分化のバランスを制御していることを示唆している。
４）Xenopus Brachyury2およびホヤBrachyuryの抗原を精製するため、FLAGタグを用いたところ、これまでのGST融合タンパク質よりも安定して大腸菌内で発現させられることがわかった。
