Outline of Annual Research Achievements

Sorting nexin (SNX) 27によるATP-binding cassette transporter A1 (ABCA1)の発現量制御をin vivoで実証するため、肝臓特異的SNX27ノックアウトマウスの作出に成功して解析を行ったが、その結果はin vitroとの乖離が見られ、これは細胞内局在の違いで説明できることを示唆する結果を得ている。
進行性家族性肝内胆汁うっ滞症1型(PFIC1; Progressive familial intrahepatic cholestasis type 1)は、ATP8B1の遺伝子変異が原因となる希少難治性肝疾患である。当研究室では、PFIC1の新規治療候補薬としてフェニルブチレートを見出しており、今年度は、PFIC1に対するフェニルブチレートの有効性、安全性を検証すべく、PFIC1症例を対象とした探索的臨床研究を実施した。本研究への参加を希望するPFIC1患者のゲノムを解析し、遺伝子変異部位を同定するとともに、当該変異を導入したATP8B1を構築しのin vitro解析を行うことにより、本研究に適切な患者を選別した。本研究内容に関しては、臨床所見と合わせて論文化した。
