Research Abstract

PCR-Select SubtractionによりJVSマウス及び対照マウス(c57BL/6J)の心臓間でmRNA発現量に差のある遺伝子を1186clone単離した.発現量の差が特に大きかった,56cloneについてシークエンスを行い,BLASTによりホモロジー検索を行った.その結果,23cloneが既知の遺伝子,15cloneが未知の遺伝子であった.今回のシークエンス結果から,JVSマウスの心臓においてup-regulateしている遺伝子にはribosomal proteinやストレス応答因子が多く含まれていることが分かった.このことから,JVSマウスでは,カルニチン欠乏により脂肪酸代謝障害が起きており,これが刺激(ストレス)となってこれらの遺伝子が増加したとも考えられる.これらはJVSマウスの心臓におけるミトコンドリアの増生(ミトコンドリアタンパク質の増加)にも関っている可能性があり,大変興味深い.
JVSマウス心筋で対称マウスに比し差の見られた遺伝子(III,IV,V,VIII)及びp53(コントロール実験のための遺伝子)のシークエンスから21bpの配列を順配列→hairpin部分→逆配列からなるsiRNAを設計作製した.これらの二本鎖DNAをpSilencer 1.0-U6 siRNA Expression Vector(3292bp; Ambion,#7207)に組み込み,C2C12へトランスフェクションした.その結果,ミトコンドリアの凝集に関わるもの,核や細胞の大きさを顕著に増大させるものなど,興味深い実験結果が得られており,今後詳細に検討する必要がある.
