Research Abstract

本年度は生体膜に結合した状態で生理活性機能を示す生理活性ペプチドの立体構造と運動性を固体NMRの手法によって観測できる磁気異方的相互作用を解析する方法の開発を行い、以下の成果を得た。
化学シフト異方性を利用した構造解析法の開発 ペプチド結合を形成するカルボニル炭素の化学シフトテンソルのδ_<33>軸がペプチド面に垂直であること、およびδ_<22>軸がC=0軸に平行であることに注目して、膜と結合してヘリックスを形成するペプチドの膜結合構造解析法を開発した1人リックスを形成するペプチドがヘリックス軸のまわりで大きな振幅の運動を持つ場合には、カルボニル炭素の^<13>C NMR線形は軸対称粉末線形になり異方性が大きく減少することが分かった。さらに磁場に配向した状態でのカルボニル炭素の^<13>C化学シフト値とマジック角で回転したMAS法で得られる等方化学シフト値、さらに低温で測定して得られる固定した状態の異方的化学シフト値を組み合わせることで、ヘリックス軸の膜面に対する配向の角度およびペプチド面のねじれ角を精度よく決定できることが分かった。
メリチンの膜結合構造の膜厚依存症 メリチンとリン脂質をモル比で1:10に混合して調製した2分子膜小胞は楕円形になり、磁場に自発的に配向することをすでに明らかにしている。今回はアシル鎖の長さの違うリン脂質、DLPC,DMPG,DPPCを用いてこれらの膜と結合したメリチンの構造を上に述べた解析法を用いて決定した。この結果、全てのリン脂質膜においてメリチンは膜に対して垂直にヘリックス軸が挿入した構造をとることが判明した。さらにN-末端とC-末端側のヘリックスの折れ曲がり角度はほぼ120度であり結晶を形成したときの角度によく一致することが判明した。しかし膜厚の厚いDPPC膜の場合ヘリックスは膜貫通する長さを持たないことが分かった。
