Research Abstract

周生期マウスに対するエストロゲン曝露によって生じる膣の不可逆的変化の分子的機構を探った。まず、エストロゲンによる上皮の異常な分化にはFGF7/FGFR2IIIbシグナル伝達系が必須であることが明らかとなった。エストロゲンは、これら成長因子-受容体そのものの遺伝子発現の誘導能を持たないが、FGF結合蛋白質の発現を誘導することで、FGF7シグナルを増強し、上皮の異常分化を生じさせる可能性が示唆された。次に、エストロゲン毒性作用に影響を及ぼすビタミンAの作用に関して、エストロゲンにより周生期特異的に発現することから関与が示唆されていたRBP2の発現解析をex vivo実験系を用いて行なったところ、in vivoにおける実験とは異なり、エストロゲンやFGF7はRBP2の発現誘導を起こさずに膣上皮の連続的増殖を生じさせた。また、ビタミンAはex vivo実験系では、エストロゲンの毒性作用を抑制しなかった。これらの結果から、第一義的には周生期エストロゲンによる膣上皮の異常分化に対するビタミンA作用およびRBP2の役割は限定的なものであり、ビタミンAは、膣以外の組織に対する間接的な作用を介して、異常分化した膣上皮が間質の性質に影響を及ぼす段階や間質が上皮に増殖シグナルを送る段階以降の後期段階に影響を及ぼし、エストロゲンの毒性作用を見かけ上抑制することが示唆された。他方、ビタミンDが、エストロゲンおよびFGF7の毒性作用の初期の段階(上皮の異常分化)を抑制することが、新たに明らかとなった。さらに、エストロゲン曝露により周生期膣組織で発現が変化する遺伝子のDNAアレイを用いた解析を通して、異常上皮の分化マーカーとして、粘膜細胞の増殖調節因子であるTFF1およびPRAPが同定され、上皮の異常増殖や癌化のプロセスに対する関与が示唆された。またエストロゲン様物質による雄生殖輸管の発生や精子の異常に対するビタミンAの抑制作用も明らかとなった。
